چکیده

iii ACKNOWLEDGEMENTS First and foremost, I have to thank my research advisor, Prof. Ronald Labbe for his guidance, encouragement and dedicated support in every step throughout the process. The accomplishment of this paper is due to his understanding and unfaltering faith in me over the last three years. While working with him, I not only developed as a professional, but also as an individual. I would also like to show my gratitude to my committee, Dr. McLandsborough for their guidance and valued ideas for my dissertation. Special thanks also to Prof. Kalidas Shetty and the food science department; particularly Ruth Witkowski and Fran Kostek for their continued support. Successfully completing my dissertation required more than just academic support, and I would like to thank my friend and fiancé Varun Rai and my friend Siddheshwari Advani for listening to, and having to tolerate me over the past three years. I would also like to thank Chi-An Lee, Kyle Landry and Chandrakant Ankolekar for their personal and professional support over the time I spent in lab. Most importantly, thanks to my family, without whom none of this would have been possible. To my parents and sister – your unwavering support gave me the strength to succeed and this dissertation is a testament to your unconditional love and encouragement. iv

Inactivation of spores of Bacillus anthracis Sterne, Bacillus cereus, and Bacillus thuringiensis subsp. israelensis by chlorination.

Three species of Bacillus were evaluated as potential surrogates for Bacillus anthracis for determining the sporicidal activity of chlorination as commonly used in drinking water treatment. Spores of Bacillus thuringiensis subsp. israelensis were found to be an appropriate surrogate for spores of B. anthracis for use in chlorine inactivation studies.

Physical characteristics of spores of food-associated isolates of the Bacillus cereus group.

All 47 food-borne isolates of Bacillus cereus sensu stricto, as well as 10 of 12 food-borne, enterotoxigenic isolates of Bacillus thuringiensis, possessed appendages. Spores were moderately to highly hydrophobic, and each had a net negative charge. These characteristics indicate that spores of food-associated B. thuringiensis and not only B. cereus sensu stricto have high potential to adhere to...

Rapid characterization of spores of Bacillus cereus group bacteria by matrix-assisted laser desorption-ionization time-of-flight mass spectrometry.

Matrix-assisted laser desorption-ionization (MALDI) time-of-flight mass spectrometry was used to characterize the spores of 14 microorganisms of the Bacillus cereus group. This group includes the four Bacillus species B. anthracis, B. cereus, B. mycoides, and B. thuringiensis. MALDI mass spectra obtained from whole bacterial spores showed many similarities between the species, except for B. myc...

The plcR regulon is involved in the opportunistic properties of Bacillus thuringiensis and Bacillus cereus in mice and insects.

Bacillus thuringiensis has been widely used for 40 years as a safe biopesticide for controlling agricultural pests and mosquitoes because it produces insecticidal crystal proteins. However, spores have also been shown to contribute to overall entomopathogenicity. Here, the opportunistic properties of acrystalliferous B. thuringiensis Cry(-) and Bacillus cereus strains were investigated in an in...

Distribution and toxin genes of Bacillus cereus spores isolated from vegetables

The objectives of this study were to evaluate the prevalence of Bacillus cereus spores in vegetables and to investigate the toxigenic characteristics of spore isolates. The detection rate of spores was 26.7% (12 out of 45 samples) which possess a higher risk of food poisoning than vegetative cell. The occurrence rate of toxin genes in spore isolates were 91.7, 100 and 100% for hblCDA, nheABC an...
